Synthesis and purification of the PeptidePeptides were synthesized by Fmoc chemistries by solid-phase synthesis as described Reference 8. Briefly, the hexapeptide was synthesized by solid phase on a pMBHA-resin (p-methylbenzydrilamine-resin) with AM-handle, which allows the cleavage of the peptide amide in acid conditions with the concomitant deprotection of the side chains protection. The elongation of the peptide chain was carried out using the Fmoc/tBu strategy. The resulting peptidyl resin was treated at room temperature with a mixture of TFA/thioanisol/H20 (95:2.5:2.5, v/v/v, 7 ml/g resin) for 2 h. The crude peptides were precipitated by filtration into cold diethyl ether and vacuum-dried. The crude product was dissolved in 10% acetic acid for de-tert-butylation at 600C and treated with DIAION for purification. Characterization was done by ESI/MS and analytical HPLC using Kromasil C8 column (4.6 x 250 mm, 5 µm, 100 Å) flow rate 1 ml/min, eluent A 0.1% TFA, eluent B 0.07% TFA in CH3CN. Elution conditions: isocratic 11 % and 0 to 40% in 30 min gradient. For further clinical data on Argireline®
The anti-wrinkle and anti-aging properties of this hexapeptideAt a 10% inclusion rate of this non-toxic, non-irritant hexapeptide molecule, is was noted that it modulates the excessive stimulation of the facial muscles, relaxing facial tension and reduces the depth of already formed wrinkles. This peptide is the main ingredient of our Créme de Vie anti-wrinkle and anti-aging cream. For more on the effect that of our Créme de Vie has on your skin and on wrinkles, please click here.
